研究方向
聚焦蛋白质凝聚体与神经分化发育及病变的机制研究,在Cell、Nat Cell Biol、Mol Cell、EMBO J、Nat Commun、PNAS、J Cell Biol等国际专业期刊发表SCI论文60篇。近期的主要发现为:1)凝聚体调控神经干细胞不对称分裂:提出相分离驱动命运决定因子时空性聚集进而介导细胞极化及干性传递的新机制;发现相分离凝聚体可突破经典复合物组装中的化学计量限制,容纳竞争性蛋白共存,以促进复杂蛋白网络动态构建。2)凝聚体调控神经元发育:发现凝聚体可介导神经元中靶标mRNA的转运与局部翻译,调控树突发育与可塑性;而蛋白质局部翻译与周转的失衡可抑制自噬通路,加剧神经毒性蛋白及脂滴的聚集。3)凝聚体决定细胞命运:发现病理条件下MBD2-NuRD凝聚体的形成抑制转录,促进胶质瘤发展,并在单氨基酸精度上验证了扰动病理性凝聚体在干预肿瘤发展中的潜力。先后主持国家重大科学研究计划课题、国家重点研发计划课题、国家自然科学基金重大项目课题等。现任中国生物物理学会理事及生物大分子相分离与相变分会秘书长,中国细胞生物学学会细胞结构与细胞行为专业分会副主任委员。
