研究方向
世界卫生组织和柳叶刀杂志报道,超过全世界人口的20%,也就是15.7亿人有听力损失,听力残疾占我国残疾总人口的24%。课题组围绕听觉医学开展如下工作:1)基于基因置换或CRISPR/Cas9基因编辑的遗传性耳聋基因治疗及临床转化;2)内耳毛细胞再生和听觉保护的研发、临床转化。发表SCI文章86篇,其中第一/通讯作者60余篇,单篇最高影响因子168.9,包括The LancetBMJ-British Medical JournalScience Translational MedicineNeuronCell ResearchMolecular TherapyNature CommunicationsGenome BiologySignal Transduction and Targeted TherapyResearchCell DiscoveryHuman Gene Therapy (封面导读)等著名期刊。取得了一系列创新性的结果,包括:与国际著名基因编辑专家David Liu、著名听觉领域专家Zheng-Yi Chen教授合作,首次以CRISPR/Cas9对活体听觉细胞进行基因编辑,发表在Nature Biotechnology;建立和发现了遗传性耳聋基因治疗内耳导入新路径和新载体;阐释了GJB2(第一大耳聋基因)SLC26A4(第二大耳聋基因)KCNQ4等致聋的分子机制;开发了基于基因编辑对Myo6KCNQ4遗传性耳聋和药物性耳聋的听力纠正;创新性地对Tmc1耳聋的RNA编辑和Klhl18耳聋的精准纠正改善了遗传性耳聋小鼠的听力;发现了成年哺乳动物内耳支持细胞和毛细胞重编程增殖再生;主导国际第一个取得疗效的耳聋基因治疗临床试验,成功纠正聋哑耳聋的听力和言语,发表在国际顶级医学期刊The Lancet(柳叶刀)。主持包括科技部国家重点研发计划课题、国自然杰青、优青、面上、青年、上海市科委细胞与基因治疗专项、上海市科委生物医药科技支撑专项、上海市科委政府间合作(芬兰)、上海浦江人才、上海市曙光学者、上海市申康临床研究项目、上海市卫健委杰青等纵向课题。近56次在权威国际大会发言。2015-2018年担任美国耳鼻咽喉头颈外科研究协会(ARO)国际委员会委员(中国代表),2018-2021年任美国ARO执行委员会委员(亚洲代表)。现任中华医学会耳鼻咽喉头颈外科学分会委员、中国生物物理学会听觉、言语和交流研究分会常务委员、中国听力医学发展基金会听觉医学创新与转化专家委员会主任委员、上海市医学会耳鼻咽喉头颈外科专科分会委员和上海市医学会罕见病专科分会委员等
